No, not if it can avoid it.  If you motion correction using 'realign'
without reslicing, then SPM only changes the image affines, and
doesn't reslice the images.

If you then do spatial normalization and select the realigned (not
resliced) data - the whole transformation is applied in one go (motion
correction + spatial normalization),
