Is there anything relevant in /var/log/messages?

How is Speakup configured - which synthesizer? Are you using
Speech-Dispatcher with Speakup, Espeakup, or a serial synthesizer?

I've had this all working fine with Espeakup, but I am not a regular Speakup
user.
